Since the movie is coming out in two months, maybe we can put all spoilers in this thread. It's cool to comment on the spoilers but if you post a spoiler yourself, please include your source and proof so this thread doesn't fill up with made up stories.
So I'm going to kick this thread off with some movie spoilers that were tweeted to me yesterday. A while ago, I tweeted Minae Noji and asked her what origin they were going with for Karai. She didn't answer me but yesterday, a guy who played a foot soldier in the movie responded to that tweet and then proceeded to give me some more juicy info. So I'm just going to post my questions and his answers and you can take from it what you will.
NOTE: The following contains spoilers. I did not initiate this conversation. This guy reached out to me so I asked him a few questions all out in the open on twitter. Feel free to check out the real conversation on my twitter page. However, you might want to stay off his page. There are no additional spoilers and several X-rated photos you can't unsee. He seems honest and legit, and I'm assuming he has permission to answer these questions on a public forum like twitter. I have no knowledge of his legal standings with Paramount.
Me: What origin are they going with for Karai
Foot Soldier: oh she is Shredder's Daughter. Also there's a theme in the Ninja Turtles Movie, the Turtles r named after famous painters so in scenes the Foot Ninjas r tied up to resemble their painter's art Example: in the subway scene we r tied up as Leonardo Di Vinci's: (The Last Supper painting) I was Phillipus (Imagine sitting on ur knees in full gear while tied up 2 hours straight!) if I was (Dirty joke about Megan Fox that I won't repeat here) tied up on my knees 2 hrs that'd be cool, but just sittin there was terrible. that's not to say @minaenoji didn't do any of her own stunts, in that subway scene she did do at least 1 of her own stunts. I'm not gonna say what she did, u'll have to see the Movie, as much as I am mad @ParamountIRL 4 not paying me, @minaenoji @ParamountIRL I just wanted to let everybody know Ninja Turtles 2014 had A LOT of thought & creativity in it.
Me: What about the Turtles' backstory?
Foot soldier: not aliens if that's what u think, genetically altered mutants it actually says so in the trailer. I had to wait to release all this info (plus most of it i probably shudn't have) but they do listen to the fans.
Me: Oh yes I know that but what about Hamato Yoshi and Oroku Saki?
Foot soldier: Splinter i wasn't still on set when they decided him, but shredder is William Fitchner April O'neil's boss at the news station. but u don't find that out until Will invites April (Megan) to his castle & it's an ambush for the turtles (it was a real castle
Me: I thought that was Whoopi Goldberg
Foot soldier: sorry i was dropped b4 Whoopi came, she is Megan' boss, I forget how Will knows April in this movie, but he takes her to his house which was a Castle, I was rarely in scenes this location: i shot this stuff May & June 2013 so it's hard to remember hold on while i look up the castle
Me: I think a lot of fans are curious about the turtles origin not staying true to the comics, no Hamato Yoshi, and if the turtles are quiet ninjas or loud and in the open.
(The part that makes me cringe a bit...)
Foot soldier: sewer mutants? no. Aliens? no. well obviously they're ninjas, do I personally know all the info? hell no I was fired in June 2013, which is y I can say any of this, but I'm not gonna guess randomly, watch the damn Movie u'll enjoy it! but honestly I grew up a fan, fans gotta understand, f*#k THE "Origin" we wund't have had Dark Knight being strict on what was the original concept, I don't go to the movies to watch the same movie i saw 10 years ago, it's not Jurassic Park revamps. I can only tell u what I know cuz @ParamountIRL fired me illegally & broke my contract, so I can say whatever here 1 thing. Megan Fox ( @meganfox ) is really that hot in person, she's obviously never had plastic surgery.
Me: Well I hope the fans like this. I would like to see other characters like Krang, Bebop and Rocksteady. Maybe we'll see them in a sequel?
Foot Soldier: I hope so! just know they do listen to the fans suggestions (They talk about it a lot on set), the fans voice is never ignored
So it's pretty clear this foot soldier hasn't read the script so he didn't have a lot of information. Still it's interesting that Eric Sachs lives in a castle. I'm assuming that is where he and April O'Neil were talking in the trailer. It's also pretty clear that the turtles are created in a lab. (IDW maybe). I found it cool that the filmmakers were discussing fan suggestions on set. That is probably what Fichtner was talking about when he said "We changed things as we went along". Also interesting is Karai being Shredder's daughter in this, even though she is asian. I'm wondering how that all connects. I'm pretty excited for this movie.

There's so far you can push suspension of disbelif before it brakes.
Also, Shredder news editor? that seems..... odd.

the Foot S seemed like he was lying to get fans to watch the movie. first he said shredder was the boss of April then he corrected him and said woopy was wtf LIEING.

Well, in Foot Soldier's defense, according to him he as sacked before Whoopi was signed on. Now, I have flashing images of Burne as Shredder... It's like JJ being teh Green Goblin

I don't think he was "lying". He was playing a foot soldier so he probably never got to read the actual script. Everything he told me was just stuff he picked up from being on set. It makes sense that he wouldn't have all of the information. Also like he said, he was fired over a year ago and probably doesn't follow the movie as closely as us hardcore fans do.

Yeah probably. At least that is consistent with past incarnations. (minus the race change.)

Wait, April will be fighting on her own too? And knowing Martial Arts? I know for a fact, In some incarnations, Splinter taught April some Martial Arts, As evident in the Nick series, But for the movie, Do they do that as well? Well there be scenes in the movie where Splinter teaches April a few martial arts moves she can use to defend herself? Plus it be kind of funny seeing Megan Fox doing Martial Arts, But that would mostly be done by a Stunt Double anyways.

Ha oh man.
Man oh man I hope this is true.
So, if true, it's possible they've effectively wiped out any direct Japanese origin for Shredder and Splinter/Yoshi?
My question here: is Splinter a reincarnation of Yoshi a la the IDW comics? Or is he "genetically enhanced" to know martial arts somehow, what with the "genetic tampering" referred to with the Turtles?
From a pure representation perspective, it's hard for me to stomach the idea of whitewashing Shredder and potentially erasing Hamato Yoshi, no matter what the reasoning.
(Not to presume any of this is true, mind you.)

If he believes that, I've got a chain of islands in the Bahamas to sell him, for a very reasonable price.

You know, I kind of like the whole "genetically splicing" using the ooze angle. It's just as believable as the other origins, and even though its different, it makes sense as it feels like a modern updated version that will probably make more sense to today's audiences. It even sort of honors every origin story so far. It honors the animals becoming humanoid with ooze like in the comics. Honors the OT series where you half mutated into the last thing you touched. (you could say that was just a kid friendly version of DNA splicing.) And it also honors the IDW series where Splinter and the Turtles start out in a lab and are the result of experiments. There's no denying now that the origin is certainly borrowing more from the IDW series than anything else.
I'm cool with April knowing martial arts because she knows martial arts in several of the different incarnations. I'm stoked about Krang being in this. To me it sounds like they've made the ultimate turtle movie. They took from every incarnation. Yes elements were changed but just based on the few spoilers we've heard all ready, I'm confident as a turtle fan that I will like this movie.

if he was just a foot extra its highly plausible the dude never got anything resembling an actual script
only directors directors main actors and a select few are given full scripts
rarely are extras given anything with dialog for this very reason...on set one day gone the next,,
im taking all this with a grain of salt...if its on screen then most of my fears are true if its not then maybe those changes they promised actually happened

Not really a spoiler but from the toys..
They were born in the shadow they live in the shadow. and they fight in the shadow. New York City. Crime rates are on the rise and innocent civilians tremble in fear. But there's a force fighting back, vigilantes among us ready to protect the city. Are they hereos we expect them to be? And are they powerful enough to take down the masked warrior everyone fears.....The Shredder?
I'm assuming that Karai would be his adopted Daughter, All past incarnations of Karai had her being adopted by the Shredder.Only the 2003 and 2012 incarnations have been his daughter.

Not really spoilers, just a theory.
There is a scene in one of the first trailers where Eric walks up to what we all assume is his empty Shredder outfit. Well, take a closer look at that scene, as I believe it isn't what it seems. I believe Eric Sachs is a character to just throw us off and anger us to help drive word of mouth; I believe Eric Sachs is taking orders from Shredder, and Eric is just a face Shredder uses to speak with the public in New York city. Is it the Saki shredder we all know and fear? Who knows.
Another thing to point out, Splinter and Shredder in this film heavily carry Japanese Motif in their design It would somewhat be in bad taste to add all that for the sake of look and then apply it to a bunch of Caucasian characters.
Still not convinced? Well, I have more to go on. One of the big reasons Shredder wears his armor has a lot to do with the damage done to him by Master Splinter, and of course, as a way to intimidate and cause fear in civilians or any one that tried to get out of underneath the power of his Foot Clan. It would seem weird for the shredder armor to exist just because and not for similar reasons that we are used to.
So to sum it all up, I believe that Eric Sachs is just someone that Shredder uses to push the ideals of the Foot clan in New York. I think the character is there to also anger fans, so they can drive the film in word of mouth, regardless if its positive word of mouth or not. It always tends to be alright as long as people are talking about said movie. I believe we are being thrown the old red herring.

I've been on this same wavelength for awhile now. I think it probably lends credence to Sachs being a Utrom and Shredder being a Krang type of alien. It's hard to tell with what little footage we've been given, but we haven't seen much to indicate that it is Fichtner in the suit other than what we've been told. It's only been implied. And I think Fichtner even said something along the lines of Shredder not being who we think he is.
