Donnie is the brains and thus the true leader. Everything Leo does is gonna be informed by Donnie, basically the most trusted advisor. He knows all the answers to the why, who, and how of every op. Thus he has more power than Leo, effectively. The other 3 Turtles might be able to fight ninjas and other basic toughs like champs but as soon as any kind of unsolved mystery or tech gap comes into play they are gonna get nowhere without Donnie!

I don't find Leo compelling as a leader, and its understandable that he was eventually replaced by Raphael in the later iterations (e.g. Raphael: Mutant Apocalypse, Rise of the TMNT etc.). Leo is neither here nor there and doesn't demonstrate the assertiveness or confidence to be the rightful leader. He desperately wants to be Splinter but keeps coming up short. He's also weirdly hesitant and neither humble nor proud in situations. Leo's not exactly neutral as he makes peculiar decisions, almost spontaneously, without rationale (as opposed to Don). He's also very easy to manipulate via potential love interest, be it Karai or Lotus (I suppose the same can be said of Raph and Mona or Mike and Renet, to a lesser degree).
Mike has the most natural talent (albeit wasted talent due to his laziness and general apathy), he's the best driver, the most agile, and perhaps, strongest turtle (later interpretations have him as non-viable and derped out). He's the stoner turtle that ironically became the face of the TMNT during the peak of America's anti-drug crusade.
